嗅球廣泛投射麩胺酸神經纖維到杏仁核(amygdala),因此嗅球切除手術會導致麩胺酸神經系統之功能發生改變,這種改變可能參與嗅球切除之大鼠所出現的異常行為。

本論文第一部分之實驗以[3H]MK-801受體結合分析法探討大鼠經嗅球切除後,其各腦區內N-methyl-D-aspartate(NMDA)受體性質之變化。

實驗結果顯示,嗅球切除會導致杏仁核及大腦皮質內之NMDA受體密度降低,且紋狀體內NMDA受體對[3H]MK-801之親合力下降。

在第二部份之實驗,為釐清NMDA受體在攫殺小鼠行為中所扮演之角色,對嗅球切除之大鼠給予全身性投予NMDA受體之拮抗劑MK-801,或將MK-801以微透析探針局部灌流到內杏仁核(medialamygdala)。

實驗結果顯示,全身性或局部阻斷內杏仁核裡之NMDA受體,均會抑制嗅球切除之大鼠表現攫殺小鼠行為。

第三部份之實驗,以開放空間行為測定儀及微透析技術,探討嗅球切除之大鼠在新環境中所出現的過動性及紋狀體內麩胺酸釋放量之變化情形。

結果顯示新環境之壓力會導致嗅球切除之大鼠其紋狀體內之麩胺酸釋放量增加,而且合併出現過動性。

本論文第四部份之實驗係以ibotenicacid破壞基側杏仁核(basolateralamygdala),結果發現嗅球切除之大鼠在新環境中之過動性及麩胺酸神經系統過度活化之現象會受到抑制。

結論:杏仁核及紋狀體內之麩胺酸神經系統參與嗅球切除之大鼠所表現之攫殺小鼠行為及其在新環境中之過動性。

Olfactorybulbectomy(OBX)transectstheglutamatergicfibersprojectingfromolfactorybulbstotheamygdala;itcausesthechangesofglutamatergicactivityandconsequentlyresultsintheabnormalbehaviorsobservedinOBXrats,mouse-killingbehavioraswellashyperactivityinnovelenvironment.The[3H]MK-801bindingassaywasusedinthefirstexperimenttoevaluatethechangesofN-methyl-D-aspartate(NMDA)receptorbindingcharacteristicsfollowingOBX.TheresultsshowedthatOBXdecreasedthedensityofNMDAreceptorinthecortexandamygdalaandalsoreducedthe[3H]MK-801bindingactivitytotheNMDAreceptorsinthestriatum.Theresultsfromthesecondexperimentshowedthatthemouse-killingbehaviorinOBXratswasinhibitedbyMK-801administeredbyperitonealinjectionandlocoalperfusiontothemedialamygdala.Inthethirdexperiment,thenoveltyexposure-inducedhyperactivitywasaccompaniedbyanincreasingreleaseofstriatalglutamateinthestriatum.BoththehyperactivityandenhancedstriatalglutamatereleaseseeninOBXratsinnoveltyexposurewereblockedbytheibotenicacidlesionofthebasolateralamygdala,whichdemonstratedinthefourthexperiment.Takentogether,theseresultssuggestthattheglutamatergicpathwaysintheamygdalaandstriatumareinvolvedinthemouse-killingbehaviorandhyperactivityinOBXrats.
